Update Resource file in App_GlobalResources Folder

Hi All,
I need to update App_GlobalResources files.
Once I publish my website, App_GlobalResources folder converted to dll. Is it possible to udpate resource file after publishing?
If possible how can I update the resource file after publishing? or is there any other way to do this?
